Does it have to be assembled with drawers?

You do not have to assemble the drawers. They are one of the last steps and are not necessary for structural integrity of the table.

Can it be assembled with out the drawers?

If you are asking can just the table be assembled without that side table/drawer attachment, I think that yes, it can. I just looked at mine again, and it seems like it's just an attached part, and the art/draft table is able to stand on it's own.

Can the drawers be put on the left side for left handed users?

I think so, but I highly suggest you look for a different desk! This one is awful. :(

Can this desk hold an aquarium tank, 40 gallon?

It is a nice desk and well made but I wouldn't put a 40-gallon aquarium tank on it. I have had fish and aquariums in the past and I definitely wouldn't do it
