The antenna is connected to your router so the operating system of the computer is irrelevant

This is the antenna ONLY, it has to be connected to another wifi device (card/access point/repeater). There is no power requirements for this device, what you are asking would be for the device this antenna connects to.

My Asus Router has three removable antennas. Does it matter which one is replaced with this panel antenna?

If you router is dual frequency (2.5/5GHz), each antenna is rated for both frequency. I used middle one of the three. You can replace anyone with direct antenna. Very important, point directly toward area you want to target (straight beam like X), it is not Omnidirectional (circular beams). Good Luck.

Will this boost the weak wifi signal from the RV park that we go to that is about 400 ft away from our camper?

That is exactly what I use it for, and it works great. I have it connected to my Ubiquiti Bullet M2, so just make sure you have the correct connectors/cables to connect this antenna to the device you want to use the signal with.
