Do you have to change the rolls between sizes or does it hold 2/3 size rolls inside and pulls the one you requested from software?

It only holds one roll. If you want to do multiples you'll need the labelwriter 450 twin turbo. It can do two rolls simultaneously.

does it print USPS priority postage

Is this printer compatabile with a variety of different kinds of labels and sizes such as the freex wifi thermal printer?

Dymo makes many sizes and kinds of labels for use in the LabelWriter 450. You would have to ask the manufacturers/users of third party labels if their labels would work in the 450. My guess would be no, unless they are specifically made to be compatible with this printer.
